Cytochrome P450 monooxygenase specific activity reduction in wheat

摘要: Roxithromycin, as widely used medicine and livestock growth promoter, arouses concern because its occurrence and persistence in soil environments. However, effects of roxithromycin in higher plants are still vague. Accordingly, we hypothesized that roxithromycin-contaminated soil may exhibits ecotoxicological effects in wheat ( ). In this study, effects induced by a gradient concentration of roxithromycin stress (0.01, 0.1, 1, 10, and 100 mg·kg ) was investigated in a 7-d soil test in . Results indicated that the specific activity of cytochrome P450 (CYP450) monooxygenase was decreased dramatically with the concentration of roxithromycin in soil. The IC value was 8.78 mg·kg of roxithromycin. On the contrary, the growth related endpoints (i.e., the germination percentage, the biomass and the height), the content related endpoints (i.e., soluble protein content and CYP450 content), and the superoxide dismutase (SOD) activity failed to reveal the roxithromycin-induced effects. Further analysis revealed that the CYP450 monooxygenase specific activity reduction was enzymatic mechanism mediated, other than oxidative stress induced. We conclude that the soil roxithromycin declined the CYP450 monooxygenase activity in by the inhibition of the enzymatic mechanism. Further efforts can include, but are not limited to, investigation of joint effects induced by combined exposure of roxithromycin and the pesticides and evaluation of the similar effects in other higher plants.

Acid resistance of methanogenic bacteria in a two-stage anaerobic process treating high concentration

摘要: In this study, the two-stage upflow anaerobic sludge blanket (UASB) system and batch experiments were employed to evaluate the performance of anaerobic digestion for the treatment of high concentration methanol wastewater. The acid resistance of granular sludge and methanogenic bacteria and their metabolizing activity were investigated. The results show that the pH of the first UASB changed from 4.9 to 5.8 and 5.5 to 6.2 for the second reactor. Apparently, these were not the advisable pH levels that common methanogenic bacteria could accept. The methanogenic bacteria of the system, viz. Methanosarcina barkeri, had some acid resistance and could still degrade methanol at pH 5.0. If the methanogenic bacteria were trained further, their acid resistance would be improved somewhat. Granular sludge of the system could protect the methanogenic bacteria within its body against the impact of the acidic environment and make them degrade methanol at pH 4.5. The performance of granular sludge was attributed to its structure, bacteria species, and the distribution of bacterium inside the granule.

Modeling of specific structure crystallization coupling with dissolution

摘要: In this paper, the research framework for specific structure crystallization modeling has been proposed in which four steps are required in order to investigate the rigorous crystallization modeling by thermodynamics. The first is the activity coefficient model of the solution, the second is Solid-Liquid equilibrium, the third and fourth are the dissolution and crystallization kinetics modeling, respectively. Our investigations show that the mechanisms of complex structure formation and microphase transition can be analyzed by combining the dissolution and crystallization kinetics modeling. Moreover, the formation mechanism of the porous KCl has been analyzed, which may provide a reference for the porous structure formation in the advanced material synthesis.

Preparation of D-lysine by chemical reaction and microbial asymmetric transformation

摘要: The DL-lysine crystals from the racemization of L-lysine was treated as substrate with AS1.1009 intact cells as biocatalysts to produce crystalline D-lysine with a yield of 56.6% from the reaction mixture after simple purification. In the presence of 0.10 molar equivalent of salicylaldehyde, L-lysine racemization can be completed within 4 h in 1.0 mol/L of NaOH at 100°C. The activation energy of the processes was 62187.86 J/mol. The characteristics of AS1.1009 decarboxylase were studied. Under the conditions of pH 8.0, temperature 37°C, cell concentration 10 g/L, tween-80 0.5 g/L, substrate concentration 30 g/L, and the specific activity of up to 3840 U, L-lysine can be completely degraded by the decarboxylase for 12 h under the optimal conditions.

Topology optimization and seismic collapse assessment of shape memory alloy (SMA)-braced frames: Effectiveness

摘要: This paper presents a seismic topology optimization study of steel braced frames with shape memory alloy (SMA) braces. Optimal SMA-braced frames (SMA-BFs) with either Fe-based SMA or NiTi braces are determined in a performance-based seismic design context. The topology optimization is performed on 5- and 10-story SMA-BFs considering the placement, length, and cross-sectional area of SMA bracing members. Geometric, strength, and performance-based design constraints are considered in the optimization. The seismic response and collapse safety of topologically optimal SMA-BFs are assessed according to the FEMA P695 methodology. A comparative study on the optimal SMA-BFs is also presented in terms of total relative cost, collapse capacity, and peak and residual story drift. The results demonstrate that Fe-based SMA-BFs exhibit higher collapse capacity and more uniform distribution of lateral displacement over the frame height while being more cost-effective than NiTi braced frames. In addition to a lower unit price compared to NiTi, Fe-based SMAs reduce SMA material usage. In frames with Fe-based SMA braces, the SMA usage is reduced by up to 80%. The results highlight the need for using SMAs with larger recoverable strains.

Experimental flexural behavior of SMA-FRP reinforced concrete beam

摘要: The most critical drawback in currently used steel reinforcement in reinforced concrete (RC) structures is susceptibility to accumulation of plastic deformation under excessive loads. Many concrete structures due to damaged (yielded) steel reinforcement have undergone costly repairs and replacements. This research presents a new type of shape memory alloy (SMA)-based composite reinforcement with ability to withstand high elongation while exhibiting pseudo-elastic behavior. In this study, small diameter SMA wires are embedded in thermoset resin matrix with or without additional glass fibers to develop composite reinforcement. Manufacturing technique of new proposed composite is validated using microscopy images. The proposed SMA-FRP composite square rebars are first fabricated and then embedded in small scale concrete T-beam. 3-point bending test is conducted on manufactured RC beam using a cyclic displacement controlled regime until failure. It is found that the SMA-FRP composite reinforcement is able to enhance the performance of concrete member by providing re-centering and crack closing capability.

Grafting of PEG400 onto the surface of LLDPE/SMA film

摘要: Linear low-density polyethylene (LLDPE) was melt and blended with styrene-maleic anhydride copolymer (SMA). The blending films were then immersed in poly(ethylene glycol)400 (PEG400) at room temperature. The surface composition of the blend films was determined by Fourier transform infra-red spectroscopy coupled with a variable incidence angle attenuated total reflection accessory (ATR-FTIR). Results show that PEG400 can be grafted onto the surface of the films via esterification with SMA. The immersion of PEG promotes the enrichment of SMA onto the surface of the films. The water contact angle data show that grafting of PEG400 onto PE can greatly improve the hydrophilicity of the PE surface. These experiments show that surface functionalization of polyethylene films by blending SMA and then surface grafting of PEG is feasible, which suggest an effective and simple route for PE surface modification via blending and grafting.

Parametric study on damage control design of SMA dampers in frame-typed steel piers

摘要: This paper focuses on damage control design of SMA dampers in steel frame piers. A parametric study based on time history analyses is carried out on frame-typed bridge piers with axial-type SMA damping device. The parameters examined are design parameters of strength ratio and stiffness ratio . Seismic performance indexes on displacement and strain are investigated under three JRA recommended Level 2 Ground Type П strong earthquake motions. Design recommendations are suggested following the results of the parametric study.

Seismic responses and resilience of novel SMA-based self-centring eccentrically braced frames under near-fault

摘要: In this paper, the seismic responses and resilience of a novel K-type superelastic shape memory alloy (SMA) self-centring (SC) eccentrically braced frame (EBF) are investigated. The simulation models of the SMA-based SC-EBF and a corresponding equal-stiffness traditional EBF counterpart are first established based on some existing tests. Then twenty-four near-fault ground motions are used to examine the seismic responses of both EBFs under design basis earthquake (DBE) and maximum considered earthquake (MCE) levels. Structural fragility and loss analyses are subsequently conducted through incremental dynamic analyses (IDA), and the resilience of the two EBFs are eventually estimated. The resilience assessment basically follows the framework proposed by Federal Emergency and Management Agency (FEMA) with the additional consideration of the maximum residual inter-storey drift ratio (MRIDR). The novel SMA-based SC-EBF shows a much better resilience in the study and represents a promising attractive alternative for future applications.

Computational studies on the seismic response of the State Route 99 bridge in Seattle with SMA/ECC plastic

摘要: This paper reports a computational study on the seismic response of a three-span highway bridge system incorporating conventional and novel substructure details for improved seismic performance. The bridge has three continuous spans supported by two single-column piers and integral abutments founded on drilled shafts. It will be the first full-scale highway bridge to use superelastic shape memory alloy bars (SMA) and engineered cementitious composite (ECC) to mitigate column plastic hinge damage and minimize residual displacements after a strong earthquake. A three-dimensional computational model capturing the nonlinear constitutive response of the novel materials and the effects of dynamic soil-structure interaction was developed to assess the seismic response of the bridge in finite-element software OpenSees. Two versions of the same bridge were analyzed and compared, one with conventional cast-in-place reinforced concrete columns, and the other with top plastic hinges incorporating Nickel-Titanium (NiTi) SMA reinforcing bars and ECC. The novel SMA/ECC plastic hinges were found to substantially reduce damage and post-earthquake residual displacements in the bridge substructure, but led to larger maximum drifts relative to the bridge with conventional reinforced concrete plastic hinges. The analysis results suggested that the novel plastic hinges could lead to improved post-earthquake serviceability of bridges after intense earthquakes.

以提高SMA利用率为目的,设计了几种不同形式的SMA橡胶支座并进行了耗能试验。研究了水平位移幅值、水平加载频率以及竖向荷载等因素对SMA橡胶支座水平刚度、耗能量以及阻尼等基本力学性能的影响,结果表明,新型的SMA橡胶支座均可有效提高橡胶支座的耗能性能和回复能力,并为SMA橡胶支座优化设计提供了依据
